Scott Sinclair notched his ninth goal in 10 games to give Swans all three points at the Madejski Stadium.
The former Chelsea winger struck the winner ten minutes before half time as he rounded goalkeeper Adam Federici before slotting home.
Nathan Dyer hit the bar for the visitors while Stephen Dobbie was also denied by Federici.
Jobi McAnuff had Reading's best chance with a 25-yard free-kick which was turned over the bar by Dorus de Vries.
